Characterizing the microbiome in anaerobic sludge digesters by stable carbon isotope signature and molecular biological approaches
Microbiome in anaerobic sludge digesters at wastewater treatment plants (WWTPs) demonstrated high complexity and diversity regarding microbial community structure and metabolic pathways,of which properly functioning methanogens and methanogenic pathways are essential for good digester performance.To characterize the microbiome under different operational conditions,stable carbon isotope signature ofbiogas was used to monitor the methanogenic pathway for 6 months in 13 full-scale and pilot-scale anaerobic sludge digesters operated at different Sludge Retention Time (SRT),Organic Loading Rates and feedstock pretreatments.Molecular biological tools were used to analyze the microbial community structure,including Fluorescence In Situ Hybridization (FISH),Quantitative PCR (QPCR) and 16S rRNA gene amplicon sequencing.Results showed that,different operational strategy led to different digester performance,methanogenic pathways and microbial community composition.High SRT (> 45 d) or application of Thermal Hydrolysis Process (THP) may result in increase of NH3 concentration in the liquid phase.The high NH3 stress (> 100 mg/L) inhibited the acetotrophic Methosaetaceae and could induce process instability,but facilitated the predominance of hydrogenotrophic methanogenesis and the growth of relevant microorganisms.Stable carbon isotope signature illustrated consistent results regarding predominant methanogenic pathways with molecular biological analysis of microbial composition,thus appears to be suitable for detecting metabolic shift in anaerobic digesters and to be a complementary measure for early warning indicators of process failures at industrial scale.
