GeoChip based microbial community analysis of a full-scale temperature-phased anaerobic digestion plant treating municipal sludge

  Anaerobic digestion is a promising technology for treating high organic wastes including municipal sludge and recovering energy via biogas production.A robust microbial community is essential for overall stability of the process and is susceptible to various operational parameters.A full-scale temperature-phased anaerobic digestion plant treating municipal sludge was followed for one year to investigate the microbial community shift and their correlations with environmental variables based on a metagenomic technology-GeoChip 5.0.Significant differences in functional microbial community structure were found between the moderate thermophilic and the mesophilic reactor.Samples from the two reactors showed notable microbial community shift throughout the year especially in the moderate thermophilic one.Genes involved with metal homeostasis were highly abundant,revealing essential effect of trace metal elements for the anaerobic microorganisms.In addition,the raw material characteristics,interior characteristics and operational parameters (pH and temperature) were determined as the key variables shaping the microbial community,contributing to a total of 75.8% and 69.5% of the variance of the moderate thermophilic and mesophilic reactor respectively.
