Dynamics of substrate specific microbes in full scale AD fed with mixed organic waste

  Anaerobic digestion (AD) of organic matter for production of biogas is a promising form of waste management.Recent studies have provided insight into the composition of AD microbiomes,but long term investigation of full-scale systems still is required to investigate microbial dynamics associated with various waste types,including its effect on AD microbiome stability during co-digestion.Amplicon sequencing of 16S rRNA and mcrA genes was applied to 46 full-scale reactors co-digesting mixed organic waste in order to investigate what drives their microbial ecosystem.Operational temperature and substrates were found to have the largest effect on community structure,functional diversity and stability profiles.Furthermore,the effects of the alternating substrates on the stability of mixed AD ecosystems were assessed by periodic investigations of the microbial communities for half a year.Differential prevalence analysis of microbial communities associated with the full-scale reactors revealed the microbial fingerprint of each designated primary waste type.Furthermore,a number of taxonomic groups were identified as having a consistent abundant presence for individual waste types.Additionally,the microbial fingerprint and associated functional diversity of individual waste types could be linked to the stability profile as well as the operational conditions of the individual full-scale reactors sampled for a longer period of time.The obtained data may serve to further investigations into the overarching microbial ecology of AD,as well as provide potential new indicator organisms for optimisation of substrate composition and inoculum slurries.
