Screening digestates for phytohormone activity

  A rapid screening chemical analysis test for auxin based plant hormones based on the Salkowski method was utilised to examine a range of industrial anaerobic digestion samples.Further work was carried out to verify these results through the development of an HPLC analytical protocol.Three digestate sample types were tested including systems with food waste,agriculture/crops and sewage sludge (acid phase and thermally hydrolysed samples) based feedstocks from 6 different sites.A significant increase in the presence of the plant hormone,auxin (indole acetic acid) was seen in all the digested samples compared to the raw starting materials.Levels of auxin increased nearly 10 fold after the anaerobic digestion of food wastes.The Salkowski test results were confirmed by HPLC analysis.Whole plant bioassays using Arabidopsis thaliana were then used to evaluate the growth promoting potential of these digestates,as compared to auxin controls.
