A Comparative Study of Thermo-Sensitive Hydrogel with Water-insoluble Paclitaxel in Molecule, Nanocrystal and Microcrystal Dispersion
Localized delivery of in situ thermo-sensitive hydrogel system may provide a long, steady and effective drug level inside of the tumor tissue.To clarify the effect of drug dispersion state and scale in such system on its outcome, we compared three types of thermo-sensitive hydrogel systems (Gel)with paclitaxel (PTX) or near infra-red probe DiR in molecules (MOs), nanocrystals (NCs) and microcrystals (MCs), respectively.It was demonstrated finally that the MOs-Gel release drug so fast, while MCs-Gel too slow, in vitro and in vivo.MCs-Gel is also unstable in terms of forming precipitate.NCs-Gel system, with moderate release ratio, results in the best tumor inhibition after introtumoral injection.
