Polymerized Nucleic acids For Delivery and Targeting

  Long chain of polymerized nucleic acids with repeated sequences can be generated by rolling circle amplification (RCA).Polymerized DNA nucleic acids have been further processed for formulating DNA nanoballs for oligonucleotide delivery or for polyaptamer nanothreads as targeting moiety.First,biomimetic DNA nanoballs were designed for delivery of multiple antisense oligonucleotides (ASOs).Secondly, RCA was used to generate protein tyrosine kinase 7 (PTK7)-specific polyaptamer nanothreads as a targeting ligand of reduced graphene oxide (rGO) with doxorubicin.
