Dual functional LipoMETs with fusogenic and apoptotic properties mediate envelope-type nanoparticles to combinational oncogene silencing
We herein report on a liposomal derivative of metformin, i.e.LipoMET, which is hypothesized to take advantage of intrinsic tumor apoptosis ability of biguanide moiety as well as a fusogenic gene vehicle to achieve the combinational antitumor effects.Systemic administration of VEGF siRNA loaded targeted LipoMETPH NPs significantly down-regulated the expression of oncogene VEGF in the NCI-H460 xenograft tumor and induced ~21% tumor cell apoptosis in vivo.In comparison with common cationic control group of LipoEDAPH NPs,LipoMETPH NPs carrying siRNA led to dramatic inhibition of tumor growth, with little in vivo toxicity.
