Complexed vesicular phospholipid gels loaded with cytarabine: antitumor efficacy against glioma

The purpose of this study was to develop polyelectrolyte complexed vesicular phospholipid gels (VPGs) loaded with cytarabine (Ara-C) that enhance in vitro and in vivo anti-glioma effects.Complexation of charged VPGs with polyelectrolyte (ε-polylysine and T-polyglutamic acid) was developed by electrostatic interactions.The TEM images and physicochemical properties of polyelectrolyte complexed VPGs verified polyelectrolyte covered on the vesicular surface.Specially, compared with ε-polylysine complexed VPGs (PLL-SCS VPGs), y-polyglutamic acid complexed VPGs (PGA-ODA VPGs)demonstrated higher in vitro cytotoxicity and cellular uptake efficiency in U87-MG glioma cells,and a single dose of Ara-C loaded PGA-ODA VPGs exhibited significant in vivo antitumor effects at least 32 days.The results were speculated that there might be γ-PGA specific receptors on the surface of cancer cells.In conclusion, complexation of charged VPGs with γ-PGA is a promising delivery depot for the treatment of glioma.
