Optimization and preparation of cRGDyK mediated pH-sensitive paclitaxel liposomes using box-Behnken design method and its antitumor activities
The formulation of cRGDyK mediated pH-sensitive paclitaxel liposomes was optimized using Box-Behnken design.Results showed that the optimal conditions were: the optimal phospholipids concentration was 17.49 mg/mL,phospholipids to cholesterol ratio was3.87∶1,drug-to-lipid ratio was 8.0∶1, respectively.In the in vitro release assay, cRPSLP/PTX showed significant pH-sensitive feature and sustained release behavior.In the MTT assay, cRPSLP/PTX could dramatically increase cytotoxicity against HepG2 cell due to receptor medicated endocytosis and pH-responsive drug release.This study shows that the Box-Behnken design method could be used to optimize the prescription cRPSLP/PTX.
