Electrosprayed particles with gambogic acid loading for lung targeting delivery
Gambogic acid (GA)-loaded particles (GA-P),with uniform particle size of 7.56 ± 0.95 μm, were prepared by technique of electrospray.With tail-vei-injection to mice, GA-P and improve the uptake of GA 2.35-times to lung, with comparison of GA solution.The antitumor efficacy of GA-P,with 86.6% of targeting efficiency (Te) to lung, is detected on pulmonary carcinoma (PC) model mice.After 2 weeks administration, PC mice in GA-P group exhibit much lower degree of tumor invasion and cell lesion in pulmonary tissue,greatly recovered lung function of longer loaded swimming time, and significantly prolonged survival time, than mice in model group, GA solution (GA-S) group, and normal saline (NS)group.Pharmacokinetics studies indicate that the preferable antitumor efficacy of GA-P attribute to not only the tissue targeting, but also the low clearance, extended retention, and high bioavaiability in plasma, and more stability of GA.
