Uptake and endosomal escape of spermine-mannan and its gene transfection on bone marrow-derived dendritic cells
In a previous study, we designed a mannose receptor-specific gene delivery vehicle named spermine-mannan (SM) for macrophage mannose receptor-positive (MMR+) cells.In the present study, we confirmed the important role played by calcium in the uptake of SM/DNA complex and the endosomal escape of the complex.The result demonstrated that SM is more efficient than other vehicles are in terms of both uptake and transfection on bone marrow derived dendritic cells.This result highlights the potential applicability of SM in dendritic cell-based immunotherapy.
