PEGylated liposomes of homoharringtonine for antileukemic therapy: safety and pharmacokinetic evaluation
To improve the safety and efficiency,long-circulating liposomes containing Homoharringtonine (HHT) (LCLipo-HHT) were developed, and their chemical and physicochemical properties were evaluated.LCLipo-HHT presented adequate properties,including a mean diameter of 75.6 ± 3.2 nm, and a zeta potential of 16.9 ± 2.5 mV.LCLipo-HHT has shown much better safety than HHT solution in the elementary toxicity experiments.The increased plasma concentration as well as blood circulation time were observed after surface modification of the PEG2000 to distearoyl phosphoethanolamine (DSPE), which may enhance the pharmacological activity of HHT to promote drug efficiency.Therefore, the results of the present work suggest the applicability of LCLipo-HHT as a new and promising anticancer formulation for antileukemic therapy.
