Poly(lactide-co-glycolide) Coated Hydroxyapatite Microspheres for Prolonged Doxycycline Release after Periodontal Pocket Administration
Doxycycline (DOXY) loaded inorganic/organic hybrid microspheres composed of hydroxyapatite (HAP) microspheres and poly (lactic-co-glyeolic acid) (PLGA) shell coating was fabricated for sustained release as reported in previous studies.Recently, the in vivo release of DOXY from the hybrid microspheres was evaluated after administration in the periodontal pockets of rabbits.The present results indicated that drug levels in the gingival crevicular fluid (GCF) could be maintained above the effective inhibiting concentration (MIC) for over 7 days for both naked HAP microspheres (HAP-DOXY) and PLGA coated HAP microspheres (PLGA-HAP-DOXY), while the burst release of DOXY from PLGA-HAP-DOXY was significantly lower.The study showed great potential applications of the drug loaded PLGA-HAP microspheres for the treatment of various periodontal diseases.
