Self-Assembled Nanosystem of Disulfide Bond Inserted Prodrugs for Paclitaxel Delivery
A novel disulfide bond linked oleate prodrug of paclitaxel (PTX-S-S-OA) was designed to facilitate the rapid and differential drug release from the hydrophobic conjugates of PTX.Ester bond linked conjugate (PTX-OA) was synthesized as the non-sensitive control.these hydrophobic prodrugs could self-assembly into nanoparticles (NPs) with uniform particle size (100 nm) and high drug-loading efficiency (over 50%, w/w).PEGylated NPs of PTX-S-S-OA showed rapid and differential release of PTX triggered by redox stimulus, resulting in significantly enhanced in vitro cytotoxicity.Such a prodrug-based nanoplatform has potential to be used as promising nanomedicine in cancer therapy.
