The pharmacodynamics study of crizotinib liposome modified with mPEG2000-DSPE on S180 bearing mice
In order to predict the suppression of liposome loaded crizotinib(CIB) on S 180 tumor lines and its toxicity to kunming male mice, the CIB PEGylated liposomes were prepared and pharmacodynamics study was conducted.The results showed that with the time history, the tumor volume of mice was increased but the tumor inhibition rate of crizotinib liposome modified with mPEG2000-DSPE (CIB-PL)was 66%, which was higher than crizotinib solution significantly(p<0.05) and the toxicity was lower yet.In this present study, we presume that CIB was loaded into stealth liposomes for the purpose of long-circulating and the enhanced permeability and retention (EPR) effect, make the drugs passive targeted to tumor tissue, and increasing the in vivo antitumor activity of the drugs,decreasing CIB adverse reactions and improving its targeting to tumor tissue.
