Co-delivery of cytostatics and siRNA: synergistic therapy to cancer cells

The aim of this study was to design a long-circulating drug delivery system with siRNA and a water-insoluble chemotherapeutic drug, paclitaxel (PTX), to enhance encapsulation of GAPDH-siRNA and therapeutic efficacy of PTX by using Nano liposome technology.Liposomes were prepared by asymmetric reverse-emulsion evaporation method.The transfection efficiencies of formulations were expressed by GAPDH Enzyme Activity tests.We showed clearly synergistic suppression of GAPDH-siRNA with PTX to tumor cells in hypoxia culture environment.In allusion to the resistant cells living at hypoxia part in the tumor, this is an appropriate treatment to improve the efficiency of chemotherapy drug.
