The influence of PEG on the TAM targeting of SA-modified liposomes
Our previous study showed that sialic acid derivative (SA-ODA) modified pixantrone (Pix)liposomes obtained a good tumor treatment effect by targeting tumor-associated macrophage (TAM).To explore whether a longer circulation time can further enhance the targeting efficiency of SA-modified liposomes, we synthesized a PEGylated SA derivative of SA-PEG2000-DSPE, and constructed a long-circulating targeting liposomes with Pix loaded inside.The SA-PEG2000-DSPE modified liposomes was nano-scaled (102.2 ±2.2 nm) with an encapsulation efficiency of 98.3% ±0.6%.The pharmacodynamic results showed that the antitumor effect of SA-PEG2000-DSPE modified liposomes was weaker than that of SA-ODA modified liposomes, and the survival time of the mice treated with SA-PEG2000-DSPE modified liposomes also was shorter than that of SA-ODA modified liposomes.Therefore, a long-circulation time cant enhance the TAM targeting efficacy of SA-modified liposomes.
