Paclitaxel-Prodrug Self-assembly Nanoparticles for Bimodal Imaging Guided Combination Cancer Therapy
Recently, nanomedicine without drug carriers draws much more attentions of many pharmacists.In this study, a novel di-payload paclitaxel conjugate with high drug loading (~78%) was developed by conjugating PTX to PTX via pH-sensitive carbonate linkage and disulfide linkage (PTX-S-S-PTX).The PTX prodrug was able to self-assemble into uniform nanoparticle with mean diameter of 123.7±2.597am.In addition, we prepared a self-assembled nanoparticle with the NIR probe 1, 1-dioctadecyl-3, 3, 3, 3-tetramethylindotricarbocyanine iodide (DiR)encapsulated within the cores for photothermal therapy (PTT) and chemotherapy applications.The DiR-loaded self-assembled nanoparticles (DSN)were featured with advantages including high drug loading, stealth-like pharmacokinetics, efficient tumor passive uptake, and the capabilities to simultaneously load with imaging or therapeutic agents.
