Preparation and Characterization of Ibrutinib Phytosome First with Egg Phosphatidylglycerol

  Ibrutinib(IBR) is a potent, covalent inhibitor of Brutons tyrosine kinase(BTK), a kinase downstream of the B-cell receptor that is critical for B-cell survival and proliferation.As a new antitumor drug, ibrutinib is rapidly absorbed and eliminated after oral administration resulting on its low bioavailability.This study was aimed to develop ibrutinib-loaded phytosomes to improve ibrutinib bioavailability with sufficient safety and stability.This system consists of ibrutinib and egg phosphatidylgycerol prepared by solvent evaporation method.The result demonstrated that molar ratio of 1:1 showed satisfying properties with mean vesicle diameter of (32.0 ± 1.82) nm and Zeta potential of(8.01 ± 0.15) mV.It showed good stability after freeze thaw stability test.Analysis of FTIR spectroscopy and DSC was confirmed the presence of physical and chemical interactions between ibrutinib and phospholipid complex.Well formed and discrete vesicles of phytosome were revealed by Transmission Electron Microscopy.
