Preparation and characterization of anti-EphA10 decorated pH-sensitive siRNA-liposomes complexes
Therapeutic delivery of small interfering RNA remains a major challenge that limits its potent clinical application.In this study, a pH sensitive PEG-schiff base-cholesterol (Chol-SIB-PEG) vas modified on cationic liposomes (PSL).To enhance targeting ability, anti-EphA 10 monoclonal antibody (Eph) was conjugated to the liposomes (EPSL).The MDR1-siRNA was incorporated into liposomes to siRNA-liposomes complexes (EPSLR) through electric interactions.The electrophoresis results showed that siRNA-liposomes complexes could bind tightly and restrain heparin displacement.EPSLR was also characterized with encapsulation efficiency.
