In vitro Propagation: Protonemal Morphogenesis and Gametophyte Regeneration of Two Species of Bryum

  The features of protonemal morphogenesis and gametophyte regeneration of two species of Bryum (Bryum coronatum and Bryum recurvulum) by in vitro propagation were studied.The process of gametophyte regeneration from spores to young gametophytes was conducted in laboratory by both solid and liquid culture.The samples of spores for gametophyte cultivation were taken from some mature sporophytes, which were collected from Hebei Province and Yunnan Province.After sterilized in 75% alcohol solution, the spores were swilled in distilled water then sowed on Knobs medium either in the culture dishes (solid medium) or the conical flasks (liquid medium).The results showed that spore germination of Bryum coronatum and Bryum recurvulum were both of the Bryum-type.Germination positions of the above two species were both located inside the cell wall.The gametophytes, which produced both on chloronema and on caulonema, had seldom rhizoids occurring in the whole germinating process.One filiform protonema cracked into two with hollow cells occurring on chloronema only.Taking about 30 days and 40 days after spore sowing, the new gametophytes of Bryum coronatum and Bryum recurvulum occurred respectively.The protonema system of Bryum coronatum occurred on the 16th day after sowing in the liquid culture.Compared with the solid culturing, the hollow cells and browning cells were more familiar in the liquid culture.Meanwhile, the taxonomic significance of the protonema system and gametophyte morphology traits are discussed.In brief, the protonema development and gametophyte cultivation research provides the scientific basis that can be applied to horticultural and pharmaceutical application to large scale rapid propagation of bryophytes.
