RESEARCH ON THE CALIBRATION METHOD OF IN-SITU SEAWATER CHLOROPHYLL SENSOR

The calibration studies for in-situ seawater chlorophyll sensor were carried out by using monospecific culture solution,to discuss calibration method and how to be applied in the oceanographic research. The laboratoryexperiments that chlorophyll sensor calibrated by Thalassiosiraweissflogii were showed that the chlorophyll concentration had good linear relationship with the fluorescence signal, the correlation coefficient R2 were greater than 0.995,the slope error of each calibration experiment is less than 5%,which means that its practical for using monospecific culture solution to calibrate chlorophyll sensor. Then comparing the results of phytoplankton chlorophyll concentration between the calibrated chlorophyll sensor and traditional fluorescence spectrophotometry, results oft-test showed that when the concentration is greater than 5mg/L,there was no significant difference between two methods;if less than 5mg/L there is significant difference, which the result of sensor was greater.Maybe high temperature and illumination influence the results of fluorescence spectrophotometry. In terms of fluorescence spectrophotometry, chlorophyll sensor can evaluate chlorophyll concentration more accurately, not only can provide the unparalleled resolutiontothe dynamics of phytoplankton, but alsoprovide support for the early warning of red tide.
