A RAPID HEADSPACE GAS CHROMATOGRAPHIC METHOD FOR THE DETERMINATION OF FORMIC AND ACETIC ACIDS IN BIOMASS HYDROLYSATE
During pre-hydrolysis pretreatment,the contents of formic and acetic acids play a crucial role in the extraction of hemicelluloses,the degradation of cellulose and the dissolution of lignin.In this paper,a recommendable headspace gas chromatography(HS-GC)method is developed to measure formic and acetic acids in the biomass hydrolysate.The method is based on the “in-vial derivatization reaction of alcohols and acids,in which the formic and acetic acids are converted to corresponding esters in the HS thermostat(instead of the outside water bath).The generated esters are then determined by the GC.To get a higher detectability(i.e.,selectivity and sensitivity),factors such as the type of derivatization reagent and the conditions of sample equilibration were optimized and validated.And then the difference between the presented HS-GC method and the HPLC method was explored by determining the biomass hydrolysates of different raw materials under various pretreatment methods.The results showed that NaHSO4/ethanol was selected as the preferred derivatization reagent.The present method had an excellent reproducibility(RSD <0.87%) and a demonstrated recovery(97.1%~103.1%).The limits of the quantification were 2.02 and 2.38 mg/L for formic and acetic acids,respectively.Furthermore,the GC analysis time only required 12.4 min.The method is rapid,sensitive and suitable for measuring formic and acetic acids in the multifarious biomass hydrolysates in pulping and biorefinery industries.
