Processivity of endoglucanase EG1 from volvariella volvacea

  Phosphoric acid swollen cellulose(PASC)was hydrolyzed by EG1(CD)which is an endoglucanase lacks cellulose binding domain from volvarilla volvacea.It was observedthat a part of mass(6.5%in 50 hrs)transfer into soluble phase,and based on BCA method,the ratio of soluble reducing groups(SRGs)to insoluble reducinggroups(IRGs)was 1.7to 1.3 depend on the incubation timerange from 1-50 hrs.Furthermore,a modified bicinchoninic acid(BCA)methoud was usedto determine the number of SRGsand sensitive fluorescence measurementwas used to quantification of theend-labeled soluble reducing group(EL)which is generated byexo-initiation and IRGs generated by endo and reducing endexo-initiations,and theapparent processivity(Papp)and the processivity initiated by exo-mode(Pexo)was calculated.The results indicated that Pappof EG1(CD)is 1.74 andsimilar to the ratio of SRGs to IRGs which was measured by BCA method.Pexo is approximately 3.2%of Papp and it is muchhigher than that of the literature cite.
