Characterization and transcriptional regulation of ELOVL6 promoter by SREBP-1 in goat mammary epithelial cells

Introduction Elongation of very long chain fatty acids protein 6 (ELOVL6) is amicrosomal enzyme involved in the elongation of saturated fattyacids and monounsaturated fatty acids with 12, 14, and 16 carbonsto 18-carbon fatty acids (C18) (Green et al., 2011).Goat milk is rich in medium-and long-chain fatty acid, four fatty acid (C 14∶0,C 16∶0,C 18∶0,and C 18∶1)account for >60% of total fatty acids in milk(Park et al., 2007).Which are substrate of ELOVL6.Promoter studies revealed that the mouse Elovl6 promoter is a direct targetof SREBP-1.Two SREBP binding sites and one E-box arerequired for maxima activation by SREBP-l(Kumadaki et al., 2008).But there is limited studies on the transcriptional regulation of ELOVL6.In this study, we aim to clone the goat ELOVL6 promoter and to investigate its transcriptional regulation mechanism by SREBP-1 in goat mammary epithelial cells(GMECs).
