NUCKS1 Regulates Milk Synthesis of Bovine Mammary Epithelial Cells

Nuclear ubiquitous casein and cyclin-dependent kinase substrate (NUCKS) was identified as a highly phosphorylated protein ubiquitously expressed in vertebrates (Grundt, K., et al., 2004).NUCKS has been reported to be a key chromatin modifer and transcriptional regulator of a number of signaling pathways,including cell death, proliferation and movement (Sargent, et al., 2008).These reports show that NUCKS shares many of the important functional properties which are important in modulation of energy homeostasis, milk protein synthesis and cell proliferation.These studies implicated NUCKS in signal transduction and regulation of cell-cycle-related processes and DNA repair.Even though NUCKS 1 seems to be important to the cell cycle and cancer progression, its exact role has not been clarified.NUCKS functions as a transcriptional coactivator for mTOR-dependent gene regulation (Beiying Q, et al.,2014), but the mechanism through which NUCKS coordinates with mTOR is not well known, furthermore, the role of NUCKS in the cell homeostasis of BMEC still remains cryptic.Our laboratory work have confirmed that Met as a representative amino acid could obviously activate the Stat5 and mammalian target of rapamycin (mTOR) to enhance milk protein synthesis and cell proliferation of BMEC.Prolactin and estrogen (E)could also obviously stimulate milk synthesis and cell proliferation of BMEC.We hypothesized that NUCKS might play a role in the functions of BMEC in respond to amino acid stimulation.Here we show that NUCKS is a key positively regulator on milk protein and milk fat synthesis and cell proliferation of BMEC via activating mTOR.
