Separation and Purification of mulberry leaf γ-aminobutyric acid
In this study, mulberry leaf γ-aminobutyric acid (abbreviated GABA) was extracted from mulberry leaves using pulsed ultrasonic and microwave-assisted extraction method.The extracts were purified with column chromatography, and the purity was measured with the liquid chromatography.The results showed the best extraction conditions were : material-solvent ratio 1 (g) ∶34 (mL), microwave irradiation power at 144 W, and extraction time at 165 s.With this extraction condition, GABA yield was 0.583%.The crude GABA extracts were washed with ethanol to remove protein contaminates, and purified with S-8 macroporous resin and sephadex G-10, and 732-sulfonic acid cation exchange resin column chromatography.The final while GABA crystals had a purity of 90.3%.
