Identification of a Hemolin Protein from Actias selene Mediates Immune Response to Pathogens

  Hemolins play a key role in development and innate immunity in insects.In this study, a hemolin cDNA sequence of 1412bp in Actias selene (As-HEM) were isolated and its open reading frames open reading frames (ORFs) were 420bp.The recombinant protein of As-HEM was expressed in Escherichia coli, and anti-As-HEM antibodies were prepared.Real-time quantitative PCR (RT-qPCR) and western blot results revealed that As-HEM mRNA and protein expressions were mostly detected in the hemocyte/hemolymph, while expressed at lower levels in midgut and fat body, and much lower amounts in other tissues tested.Also, the expressions of As-HEM mRNA and protein could be induced significantly after Beauveria bassiana, E.coli, Micrococcus luteus and nuclear polyhedrosis virus challenges.Agglutination assay revealed that recombinant As-HEM could promote the agglutination of E.coli in the presence of calcium.Our results suggested that As-HEM played an important role in the innate immunity of Actias selene.
