Analysis of Differential Proteome Related Sclerotium Disease at Early Stage of Mulberry Fruits

  The fungus disease of sclerotinia is the mainly biotic stress factors which severely impact the mulberry fruits development, and do destructive harm on the productivity and quality of mulberry fruits.In this study, the molecular mechanism was discussed at pathogenic fungus infecting mulberry, by using the diseased and normal young fruits of variety Da 10 as the experimental material.The two-dimensional electrophoresis (2-DE), mass spectrometry and bioinformatics technique were used to isolate and identify the differential expression proteins.The results showed that 45 proteins were differentially expressed more than 1.5 times, in which 18 proteins were up-regulated while 27 proteins were down-regulated compared with normal young fruits through the 2-DE map images with IPG-IEF/SDS-PAGE analysis;In which, 34 proteins identified by MALDI-TOF/TOF MS were related to many physiological processes, mainly including material and energy metabolism, also involving in gene expression regulation,oxidation-reduction reaction, cellular component and plant stress responses.Subcellular localization of the identified proteins were analyzed by bioinformatics technique.The results showed that mulberry young fruits can regulate the expression levels of multiple proteins localized mainly in cytoplasm and the organelles to reply sclerotium disease infection.The novel findings about these pathogenesis-related proteins will provide valuable information to further study the pathogenesism of sclerotinia disease and disease-resistant molecular breeding in mulberry.
