Dietary Effects on Enzymatic Activity and Gene Expression of α-Amylase in Antheraea pernyi Midgut

  The digestive tract of insects exhibits multiple, diverse and unique combination of enzymes to digest food material depending on dietary content of a range of host plants.Alpha-amylase is an important class of enzyme which is required for the digestion of starch to ready energy source required for the optimal growth and development of insects.In this study, we cloned and characterized amylase gene (Ap-Amy) from Antheraea pernyi (Lepidoptera: Saturniidae).The gene length consisted of 1794-bp open reading frame (ORF) encoding 597 amino acid residues.Expression analysis of Ap-Amy by RT-PCR indicated that the gene was detected in eggs and larvae stages and mostly transcribed in midgut of A.pernyi larvae, corresponding to major place of digestion.Amylase activity and gene expression were higher in tussah feeding on Quercus acutissima than Quercus liaotungensi.This research is helpful to screen high-feed-efficiency A.pernyi varieties and provide new strategies to increase the production of tussah industry.
