Callus induction and establishment of a genetic transformation receptor system in Lilium davidiivar.unicolor
Lilium davidii var.unicolor, which originated in China, is famous for its ornamental and edible values resulting from the showy flowers and nutrient-rich scales, respectively.Furthermore, it is highly resistant to abiotic stresses, including drought, cold stress and strongly saline or alkaline soil conditions.However, L.davidii var.unicolor has a delicate stem and small flowers, currently limiting its use as a garden plant.The value and quality ofL.davidii var.unicolor would be vastly improved if other desirable traits such as big flowers could be engineered in it using an Agrobacterium-mediated method.Callus is an admirable receptor tissue for genetic modification.This study examined the optimal condition for callus induction and differentiation as well as the most suitable antibiotic concentration for selection in genetic transformation of L.davidii var.unicolor.Results indicated that the best media for callus induction from filaments and leaves were MS + TDZ 0.1 mg L-1 + NAA 0.5 mg L-1 and MS + TDZ 0.01 mg L-1 + NAA 1.0 mg L-1.The induction rate of callus was 80.63 and 77.78% , respectively.Subsequently callus were differentiated into adventitious buds in MS medium containing 0.1 mg L-1 TDZ and 0.5 mg L-1 NAA, with a maximum differentiation rate of 86.54% of all callus and 3.33 adventitious buds per callus.In addition, the callus we obtained is sensitive to common antibiotics, and the sub-lethal concentration of kanamycin and hygromycin were 100 and 20 mg L-1, respectively.The efficiency of regeneration system presented here is twofold higher than previously reported methods in this species.
