TriPleXTM waveguide-based fluorescence biosensor for multichannel environmental contaminants detection
In order to realize the multi-analyte assays for environmental pollutants, an optical biosensor utilizing laser-induced fluorescence-based detection via the binding of biomolecules to the surface of an integrated TriPleXTM waveguide chip is described. As far as we know, this is the first demonstration of using the TriPleXTM technology to fabricate the waveguide chip, distributing the evanescent excitation light into four-channel waveguide branches through the Y-junction splitter. And hence the 32 separate sensing patches were formed on the chip surface. The biosensor with the low-loss SiO2/Si3N4 waveguide configuration enables the high chip surface intensity, low patch-to-patch deviation etc., and hence benefits for the fluorescence-based biosensing applications. Moreover, the complementary metal-oxide-semiconductor compatible fabrication of waveguide chip allows for its mass production at low cost. The as-proposed biosensor was proven with high sensitivity, selectivity and reproducibility for the multi-contaminants detection in real water samples.
