Petroleum Degradation by Soil Microbial Community and Characterization of Hydrocarbonoclastic Strains

  Interest in biodegradation of petroleum hydrocarbons and derived pollutants by bacteria has been steadily increasing in recent decades.The aim of this study was to assess the efficiency of an indigenous bacterial population enriched from a petroleum polluted soil collected from a gas station in Tunisia, in the biodegradation of a crude oil type Zarzatine (ZCO) and its various fractions.Results showed that the percentage of petroleum degradation was increased during the incubation period.Maximum biodegradation of the aliphatic fraction (NAH) (54.94%) and maximum biodegradation of the aromatic fraction (AH) fraction (69.44%) were recorded after two months of incubation.These results are supported by the GC/MS analysis.Petroleum-degrading bacteria were also isolated from the soil sample (K) and characterized.Three isolates were selected as hydrocarbonoclastic strains and were affiliated to genera Pseudomonas based on 16S rRNA sequencing.These isolates were screened by polymerase chain reaction (PCR) for the presence of three catabolic genes involved in petroleum hydrocarbons biodegradation: alkane monooxygenase gene (alkB), Naphthalene dioxygenase (ndoB) and Catechol-2,3-dioxygenase (Cat23).Our results demonstrated that alkB was successfully detected in MK1 and MK6 strains.However, both ndoB and cat23 genes were never detected in all strains.
