Immunosensor Based on Screen-printed Electrodes for the Determination of Myeloperoxidase
The production and assembling of disposable electrochemical myeloperoxidase (MPO) immunosensor,which can combine the high selectivity of immunoanalysis with the ease of screen-printed electrodes,has been carried out.In the immunosensor construction,a screen-printed electrodes (SPE) was first coated by a film of multi-wall carbon nanotubes and N,N-dimethylformamide,then the gold nanoparticles (GNPs)-Chitosan homogeneous suspension has been obtained by electrochemical deposition.And then the antibody of MPO (anti-MPO) was absorbed on the surface of GNPs-Chitosan monolayer.Finally,BSA blocked the non-specific adsorption site.The modification process was characterized by transmission electron microscope and cyclic voltammetry.The factors possibly influenced the performance of the proposed immunosensor were studied in detail.Under optimal conditions,The SPE exhibited good electrochemical behavior to the concentration of MPO from 0 to 20 U/L(r=0.98501) and gave a detection limit of 0.3 U/L.Data concerning the application of modified screen-printed electrodes to the analysis of MPO in serum samples are presented and discussed.
