Identification of MicroRNAs Response to Hypoxia in the Lung of Yak by Deep-Sequencing Technology
Multiple lines of evidence now establish microRNAs(miRNAs), which are short noncoding RNAs that regulate gene expression through post-transcriptional, as key elements in this response to hypoxia.In order to further understand the role of miRNA mediated post-transcriptional regulation in high altitude adaptaion of yak, the miRNA transcriptomes of lung were profiled using deep sequencing methods.A total of 17,562,643 clean reads were obtained from a lung library, and 454 known and 68 novel miRNA candidates were identified.The top 10 miRNAs accounted for greater than 87% of the miRNAome, which were predicted to target 2 561 genes.Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analyses showed that the identified miRNAs targeted genes enriched in signal transduction and developement processes.The results would indicated that these miRNAsdirectly target important gene transcripts that coordinate metabolicreprogramming, DNA repair, apoptosis, and angiogenesis, among many other cellularadaptations to low oxygen availability.These findings revealed for the first time the global miRNA expression in yak on the Qinghai-Tibetan Plateau and enhance understanding of yak miRNA functional regulatory mechanisms to survive hypoxic insults.
