Expression analysis of abiotic stress responsive genes in alfalfa using the annealing-control-primer-based geneFishing technique
In order to identify potential candidates for acquiring stress tolerance, a new annealing control primer system was used to identify the differentially expressed genes.Alfalfa (Medicago sativa L.) seedlings were treated with various abiotic stresses such as cold, heat, salt, drought, Cu, Cd and As.Using ACP 93 and ACP 41, one differentially expressed band was identified as the same sequence, which represents a mitochondrial small heat-shock protein coding gene, MsHsp23.This band was increased in alfalfa under heat, salt and Cd stresses.Differential expression patterns of MsHsp23 were further analyzed by Northern blot.Temporal expression analysis is showed that mRNA pool was altered as early as 1 hr of treatment.Differentia accumulation of MsHsp23 under heat, salt and As stresses suggest its potential involvement in diverse abiotic stress tolerance, and thereby making a target for further molecular analysis.
