Isolation of Lactobacillus homohiochii from aerobically stable alfalfa silage
To examine the bacterial community associated with aerobic stability of alfalfa silage, wilted alfalfa was stored in a laboratory silo for 5, 10 and 60 days with and without the addition of molasses.Plate-culture and denaturing gradient gel electrophoresis (DGGE) analyses were performed at silo opening and after 3 and 7 days of aerobic exposure.Spoilage was not found in control silage stored for 60 days and molasses-added silage regardless of the storage period.The DGGE analysis indicated presence of Lactobacillus homohiochii in molasses-added alfalfa silage, but the bacterium was not cultured in de Man, Rogosa, and Sharpe medium.Liver infusion sake medium enabled isolation of L.homohiochii;hence, association of difficult-to-culture bacteria are suggested with high aerobic stability of alfalfa silage.
