A simple and Efficient Method of DNA Extraction from Formalin-Fixed and Paraffin-Embedded Tissues for Screening EGFR Gene Mutation

  To detect mutations of the EGFR gene is critical for predicting the response to therapy with tyrosine kinase inhibitors for nonsmall-cell lung cancer(NSCLC)patients.DNA extraction from formalin-fixed and paraffin-Embedded(FFPE)tissues is a key step for EGFR Gene mutation.Here we show that a simple method of DNA extraction from FFPE tissues for efficiently screening EGFR Gene Mutation.Extracting DNA from FFPE tissues of NSCLC patients using 1%Triton X-100(pH10.0)by heating at 95℃ for 30 min.DNA extracted products were used as template for amplifying EGFR exon 21 by PCR.To detect mutations of the EGFR exon 21 with PCR products as template by DNA Sequencing.Results show DNA fragments size extracted from FFPE tissues is about 250bp~500bp.About 210bp of EGFR exon 21 can be amplified efficiently from extracted DNA.Mutations of EGFR exon 21 can be detected from PCR products by DNA Sequencing.This simple and non-laborious protocol can successfully extract DNA from FFPE tissue for screening EGFR gene mutation and should facilitate the molecular analysis of a large number of FFPE tissues from NSCLC patients.
