A Novel Antitumor Drug-loaded Biodegradable Stent for the Treatment of Colorectal Cancer

  The paper presents a novel biodegradable weft-knitted stent composing double layers witii drug delivery function,which can be used for treatment of colorectal cancer.The inner layer is a tubular weft-knitted graft made from polydioxanone yarn.The outer layer is a drug-loaded electro-spun membrane.The mechanical results showed that tiie radial force of stent can reach above 3.3 cN/cm2 under compression at 50%.This value is better than commercial Wall-stent and Z-stent from market.The drug-loaded membrane was examined by SEM,FTIR and HPLC to examine the morphology,chemical structure,drug loaded efficiency and release property in vilro.The results revealed that 5-FU as the loaded drug is distributed evenly in the eleclro-spun membranes.The HPLC results revealed tiiat tiie released amount of 5-FU was greatly elevated when 5-FU-loaded percentage increased.The release mechanism was divided into two stages,i.e.a fast release profile in tiie 24 hours,and then a stable release pattern maintained.It can be suggested that proper drug concentration,fiber diameter and evenness of tiie drug-loaded PLLA fibrous membrane are very important for drug release profile.In conclusion,this drug-loaded stent may find promising clinical applications for colorectal cancer in the future.
