Study on the Genetic Toxicity & Mechanism Induced by Cadmium and Phenol in Water by Vicia Faba Bioassays

  Genotoxicity and cytotoxicity of cadmium and phenol in water were assessed by using the Vicia faba bioassays.The relationship between cell micronuclei and lipid peroxidation induced by cadmium (1~100 mg/L) and phenol (0.5-50 mg/L) was also investigated.As a result,it was found that cadmium and phenol both induced micronuclei (MN) and pycnosis formation in Vicia faba root tips.The MN frequency and pycnosis increased significantly in the Vicia root cells under exposure to cadminum and phenol in the concentration range of 5-100 mg/L and 2?50 mg/L,respectively.The mitoticindex (MI) decreased in a concentration-dependent manner and significant differences were found in the Vicia faba roots among the treatments and the control after exposure to 5-100 mg/L cadmium and 2~50 mg/L phenol for 12h.Linear correlative relationships were identified between MN frequency and lipid peroxidation in Vicia faba roots induced by cadmium (R2 = 0.94),and phenol (R2 = 0.95),indicating that both cadmium and phenol exposures could induce oxidative stress,cytotoxicity and genotoxicity in plant cells.The Vicia faba root MN assays were thus proved to be simple,efficient and reproducible methods for the genotoxicity monitoring of water environment regarding cadmium and phenol.
