Design Orthogonal Experiment to Find the Optimal Extraction Conditions of Xylaria nigripes Polysaccharides and Evaluate Its Capacity of Antioxidant
This paper chiefly discusses the antioxidant ability of XNP.The main active substance of Xylaria sp,namely polysaccharide,can be obtained through extraction and purification.XNP is our target extract,so the orthogonal experiment is designed to discuss the optimal extraction condition of XNP.In the orthogonal experiment,three factors have been analyzed,including the extraction time,auxiliary supersonic frequency and solid-to-solvent ratio.The orthogonal experiment shows the optimal extraction condition is such that the extraction time is 90 minutes,the supersonic frequency is 80 kHz and the extraction solid-to-solvent ratio is 1:110.In the antioxidant ability experiment,the IC50 value of XNP is 695 μg/mL in the Fe2+ chelating ability experiment; the IC50 value of XNP is 597 μg/mL in the ABTS free radical scavenging experiment; and the absorption value of XNP is 0.046 when the concentration of XNP is 2 mg/mL in the reducing ability experiment.In the above three antioxidant ability experiments,with the rise of XNP solution concentration,there is a positive liner relationship between solution concentration and antioxidant ability.The above results suggest that XNP has antioxidant ability,and it will be used as one of the sources of antioxidants for making cosmetics.
