Antioxidant and Cytotoxic Activities of Pleurotus ostreatus, P.sajorcaju, Agaricus campestris, and A.bisporous in Different Solvent Extracts

  Mushrooms are known to contain a wide array of compounds with high nutritional and medicinal values.We report here a comparison of different solvents to extract antioxidant and antitumor compounds from P.ostreatus,P.sajorcaju,A.campestris and A.bisporous.Freeze dried fruiting bodies of these four species of mushrooms were extracted by seven different solvents including water,ethanol,ethyl acetate,acetone,chloroform,hexane and petroleum ether.The extracts were analyzed for their antioxidant activities using the % DPPH (2,2-Diphenyl-1-Picrylhydrazyl) scavenging activity method.Our results revealed that the water extracts exhibited the highest % DPPH scavenging activity in comparison to all other solvent extracts.The highest was from the water extract of P sajorcaju (78.1%) and the lowest was from the hexane extract of A.bisporus (0.8%).In general,extracts from non-polar solvents exhibited much lower antioxidant activities than those from polar solvents.The cytotoxic effects of these extracts were evaluated using Larynx carcinoma (HEp-2) and breast carcinoma (MCF-7) cell lines.The IC50values after treating Hep-2 cells with the hexane extracts of P.ostreatus,P.sajorcaju,A.campestris,and A.bisporous were 1.7±1.56,2.1±2.82,4.4±1.71,and 2.2±1.34 ig/mL,respectively.The hexane extracts from all these mushrooms also had similar ICs0values against the MCF-2 cancer cells,suggesting that it is the most effective solvent in extracting the anticancer compounds from mushrooms.Extracts from solvents with nonpolar or intermediate polarity are much more potent than those from polar solvents in their cytotoxicity against cancer cells.In addition,the same solvent extracts from different mushrooms possessed varied degrees of cytotoxicity,indicating the possibility that there may be a wide diversity of anticancer compounds in these four mushrooms.
