Effect of Extracts from Flammulina velutipes and Pleurotus eryngii Fruiting cBodies on Foam Cell Formation of Human Acute Monocytic Leukemia Cell Line
Atherosclerosis is a chronic inflammatory disease.Decreasing the formation of foam cells has been thought to be a potential means to prevent atherosclerosis.In this study,we used the extracts of two edible mushrooms,Flammulina velutipes and Pleurotus eryngii,to evaluate the effect on foam cells formation and related proteins expression.Based on the results of Oil red O staining,FvW,PeW,FvEA and PeEA could significantly reduce the ability of macrophage on uptake of oxidized low density lipoprotein (oxLDL) to 88.12%,88.23%,87.50% and 85.43%,respectively (p<0.05).The expression of CD36 was significantly reduced by FvEA and PeEA,but was up-regulated by FvW and PeW.The relative expression of peroxisome proliferator-activated receptors alpha (PPAR u) and liver X receptor alpha (LXR α) was up-regulated by all extracts,but PeW and PeEA had no significant effect on ATP binding cassette transporter Al (ABCAl) expression.Although FvW,PeW,FvEA and PeEA could promote the expression of PPAR α and LXR α,ABCAl was not expectedly up-regulated by all extracts.The extracts might contain some components,which could influence the cellular genetic regulation and protein expression,and thereby,the promotion of ABCAl expression was interfered.In conclusion,we demonstrated that the differentiated THP-1 macrophages on uptake of oxLDL could be effectively inhibited by extracts from F.velutipes and P.eryngii,and therefore the formation of foam cells could also be reduced.For application in prevention of atherosclerosis,the separation and purification of extracts and further active components identification are the areas of further investigation.
