The Growth Requirements of a Medicinal Fungus Xylaria nigripes
The Xylaria nigripes (Xn) is origin to mainland China and Taiwan.It is a species of ascomycetous fungi commonly found growing on abandoned termite nests.It is used frequently as a medicine in mainland China.In recent studies,Xylaria nigripes is considered to be able to treat mental illness,nervous system diseases and digestive diseases which were frequently seen in older people.Solid state culture is considered to be an economic way for commercial production of medicinal fungus with higher contents of physiological active compounds comparing to the liquid state culture.This technique has been applied very successfully in culturing Antrodia cinnamomea,a very precious medicinal fungus only found in Taiwan.The present experiment is aimed to find out the growth requirements,namely the suitable cultural medium and environmental conditions for Xn which will be very useful in commercial mass production of Xn on solid state medium in the future.Presently,Xn was found grown better in 25 ℃ with a pH ranging from 5.5~6.0.Fructose and glucose were ideal as the.carbon source,malt extract and casein hydrolysate were ideal as the nitrogen source.Nicotinic acid is beneficial for the growth of Xn.These results were used for the establishment of liquid culture which would be used as the inoculum for solid state medium culture of Xn in the future.
