Anti-Oxidant and Anti-Diabetic Effects of Mycelium and Fruit-Body Extracts of Mycoleptodonoides aitchisonii

  Mycoleptodonoides aitchisonii (Berk.) Maas Geest is a culinary mushroom that is recognized as both a nutritious food and an excellent source of bioactive compounds.The purpose of this study was to investigate the antidiabetic and antioxidant properties of M.aitchisonii both in vitro and in vivo.To measure the antioxidant activity ofM.aitchisonii extracts against peroxynitrite,peroxyl radical and hydroxyl radical,scavenging activities were evaluated by total oxyradical scavenging capacity (TOSC) assays.The results revealed that fruit-body extracts had 0.9,3.7,and 1.6-fold higher activities relative to the mycelium extract.For an in vitro anti-diabetic assay,the Akt phosphorylation method was used,which is based on the signal transduction pathway of diabetes.Fruit-body extracts significantly inhibited the decrease of Akt phosphorylation by IL-6,but there was no effect from the mycelium extract on Akt phosphorylation.At a maximum fruit-body extract concentration of 100 μg/mL,the phosphorylation level of Akt increased to 77%,whereas the level was 50% for the insulin-only treatment group.M.aitchisonii treatment also resulted in a significant reduction in blood sugar and cholesterol in a streptozotocin-induced diabetic mouse.The M.aitchisonii fruit body can potentially be used to prevent various diseases such as sugar and lipid metabolism disorders.
