Fibrinolytic and Anticoagulant Enzymes of Selected Medicinal and Culinary Mushrooms
Cardiovascular diseases are a major cause of death across many countries and are mainly caused by atherosclerosis.Thrombolytic agents are used in the treatment of thrombosis.However,a number of them have undesirable side effects including bleeding.Several mushrooms have been studied for their thrombolytic or fibrinolytic activities.The enzymes can be extracted from fruiting bodies or mycelia grown in either submerged or solid substrate fermentation.In this paper the fibrinolytic activities of the fruiting bodies of Auricularia polytricha (Mont.) Sacc,Lignosus rhinocerotis (Cooke) Ryvarden and Pleurotus giganteus (Berk.) Karunarathna & K.D.Hyde is presented.All three mushrooms studied had varying degrees of fibrinolytic activities.Auricularia polytricha gave the highest fibrinolytic activity.The effect of processing method on fibrinolytic activity was assessed using fibrin plate assay and Folin-spectrophotometric method.The fruiting bodies were either oven dried or freeze-dried.The aqueous two-phase system (ATPS) was used to further concentrate and recover the fibrinolytic enzyme/s.The parameters such as poly (ethylene glycol) (PEG) molecular mass,concentration of PEG as well as salt,concentration of crude and system pH were optimised to recover the fibrinolytic enzymes.The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was performed to determine the molecular weight of fibrinolytic enzymes.The freeze-dried A.polytricha fruit bodies had higher fibrinolytic activity of 3.88 U/ml when compared to oven-dried fiuit bodies with 2.19 U/ml activity.Poly (ethylene glycol) (PEG) 8000-phosphate ATPS was found to be suitable for the primary recovery of fibrinolytic enzyme/s from the extract.The ATPS comprising a volume ratio equal to 1.5,PEG 8000 20.0% (w/w),phosphate 11.6% (w/w),system pH of 7.0 and loaded with 25% (w/w) of crude extract from freeze-dried A.polytricha fruit bodies gave a 7.01-recovery fold.The molecular weight of the enzyme was estimated to be 66 kDa.Six mushroom namely G.lucidum,Cordyceps militaris,L.rhinocerotis,P.giganteus,P.floridanus and A.polytricha were screened for their anticoagulant activity.Only A.polytricha and L.rhinocerotis had anticoagulant activities demonstrated as antiplatelet aggregation activities.The crude extract of L.rhinocerotis was dialyzed and acetone precipitation of protein was performed to concentrate the protein in the extract.Anti-platelet aggregation activity was shown by the 45-50kDa protein fraction.Further,ATPS recovery of the protein confirmed that a protein was responsible for the anticoagulant activity.
