Activity of Cordyceps militaris Water Extract in the Cells of Yeast Saccharomyces cerevisiae
Entomopathogenic fungi of the genus Cordyceps,such as Cordyceps militaris,are used in traditional Chinese medicine and as a nutritional supplement.Extracts of Cordyceps sp.exhibit various therapeutic and pharmacological activities including immunomodulatory,anti-oxidant,anti-inflammatory,anti-tumor,anti-metastatic,hypolipidaemic and hypoglycaemic activitiy and are known to increase athletic endurance and strength.However,their exact mechanism of action in the cell still remains unknown.In the present study we investigated Cordyceps militaris water extract (CMWE) in the cells using yeast Saccharomyces cerevisiae,which is an appropriate model organism for investigating fundamental cellular processes,such stress responses and metabolic pathways.Yeast cells in the stationary phase were exposed to CMWE.After defined time of incubation (2,4 and 6 hours) at 28℃ and 220 rpm samples were taken to measure intracellular oxidation,cell energy metabolic activity and CO2 production.Results showed that CMWE increased cellular energy metabolic activity and CO2 production in a time dependent manner,but on the other side decreased intracellular oxidation compared to control cells was observed.This might be related to accelerated oxygen utilization with less reactive oxygen species production during mitochondrial respiration or to pro-antioxidative activity of CMWE.Thus,understanding of mechanism of action of Cordyceps nilitaris extract in the cell is important for its further potential application in the prevention and treatment of age-related and other diseases as well as in improving body energy performance.
