Research on the Extraction ofLentinula edodes Nucleic Acid by Enzyme with Ultrasonic Wave

  The one-dimensional laboratory method was applied to screen the best enzyme preparation for the sporoderm-broken of Lentinula edodes cells and hydrolyzation of Lentinula edodes extracting solution.The orthogonal experiment was applied to optimize the best process conditions for the extraction of Lentinula edodes nucleic acid by enzyme with ultrasonic wave and the hydrolyzation of Lentinula edodes extracting solution by.enzyme.The edible fungus hydrolase is the best enzyme preparation for the sporoderm-broken of Lentinula edodes cells,its usage is 0.5% less than the traditional one.Its A01 enzyme usage is 0.7%,A02 enzyme usage is 0.5%,with the time of 3.5 h and 5 h respectively.The ratio of sample to solution is 1:40.Then extraction samples were treated by ultrasonic wave under the power of 500 W,frequency of 20 KHz and the duration time of 30 min.The enzyme bromelin could greatly improve the nucleic acid extracting rate and purity with enzyme usage of 0.2 mg/mL,duration time of 3 h,pH value of 7.0 and temperature of 45 ℃.Under the best process condition,the Lentinula edodes nucleic acid extracting rate ranges from 3.047% to 3.195%,with the purity from 74% to 75%.In this paper,we preliminarily established a new Lentinula edodes nucleic acid extracting process,which could be applied in the healthy and flavoring food production from edible and medicinal fungus.
