The Optimum Submerged Culture Condition of Tremellafuciformis Berk.and Its Antioxidant Properties
Tremella fuciformis Berk.is a natural edible and medicinal mushroom commercially available in Taiwan.In this study,the effects of initial pH,incubation time,and various medium in fermentation fluids to optimize exopolysaccharide (EPS) production by T.fuciformis were evaluated in shaking flasks and bioreactor.The antioxidant properties were investigated as well.Results showed that cultivation in potato dextrose broth (PDB) medium at pH=9 in 48 hour-incubation time in shake flask was the most efficient condition to obtain maximum EPS.However,the quantity of EPS from various initial pH values (5-9) was not significantly different.Total polyphenol and β-glucan contents from each EPS sample (pH 5-9) were evaluated.T.fuciformis polysaccharides could remove 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical by 40.63% (10 mg/mL,pH=9).The chelating abilities of ferrous ion of all extracts reached over 80%.EPS extracted from the initial pH value of 9 showed the highest reducing power ability among other pH values (ECs0 =5.97 mg/mL).The EPS from T.fuciformis presented non-cytotoxic to the skin fibroblast of mouse (NIH/3T3) and the survival rates were more than 100% by using MTT assay.The samples were used to analyze the scavenging activity against oxidative damage induced by UVB radiation and H2O2.The results showed the antioxidant activity was in dose-dependent manner (p < 0.05).Furthermore,these findings have revealed that the EPS of T.fuciformis by submerged cultivation possessed antioxidant properties and can be used as an alternative resources for cells protection or treatment.
