Effects of Vitamins and Amino Acids on the Mycelia Growth of an Isolated Strain of Xylaria Species

  Xylaria nigripes (Koltz.) Sacc.,also known as Wu Ling Shen,is a species of ascomycetous fungus belonging to the family of Xylariaceae commonly found in abandoned termite nests.A strain of Xylaria which grown in abandoned termite nest from vegetable garden in Wuta,Pingtung,Taiwan was isolated and identified as Xylaria sp.The aim of this study was to investigate the effects of vitamin and amino acid on the mycelia growth of Xylaria sp.The mycelia growth of all the media by adding to vitamins were significantly faster than the basal (control) medium.The mycelia growth of the medium containing nicotinic acid was 1.81 times faster than that of the basal medium,in the meanwhile,the medium containing biotin,vitamin B6,vitamin B 1 and vitamin B2 was 1.43,1.36,1.23 and 1.15 times faster than that of the basal medium,respectively.In the 20 experimental amino acids,the suitable amino acids for mycelia growth of Xylaria sp.were arginine,aspartic acid,cysteine and glycine,which were significantly faster than the basal medium.Particularly,the most suitable amino acid for mycelia growth was arginine,followed by glycine,aspartic acid and cysteine in order.The mycelia growth rate of them was 1.80,1.76,1.57 and 1.52 times faster than that of the control,respectively.Based on the results in this study,the addition of some vitamins and amino acids,especially nicotinic acid and arginine,respectively,used in the medium can be considered practical and feasible for mycelia growth of Xylaria sp.
